This homogeneous fluorescence-based assay pre-loads cells expressing a GPCR of interest that signals through calcium with Rhod-4 NW, which can cross cell membrane. Non-specific esterases within the cell cleave the lipophilic blocking groups on the Rhod-4 NW, producing a fluorescent dye that stays inside the cell. Upon binding to calcium, the fluorescence of this negatively charged product will increase with increasing levels of calcium. This allows us to monitor GPCRs and calcium channels indirectly. The assay has high sensitivity, long wavelength, and more than 250 times fluorescence increase when read with excitation at 540 nm and emission at 590 nm.
