The antibody (also known as CAD; CALD1; CDM; L-caldesmon; Non-muscle caldesmon) reacts with caldesmon polypeptide in immunoblotting assays. The antibody, however, never cross-reacts with caldesmon (non muscle) or the skeletal and cardiac muscle. It exhibits specificity towards smooth muscles in immunohistochemical staining. Moreover, it does not stain the connective tissue or endothelial fibroblast cells while staining the visceral and vascular smooth muscle cells. The myoepithelial component present in the galactophorous sinuses get stained for both the normal and malignant tissues of breast. But, it may remain non-reactive when immunocytochemical methods are applied for the cultured cells of smooth muscles. This possibly demonstrates the caldesmon down regulation which is noticed in generally in tissue culture.
